The CellTiter 96® AQueous One Solution Cell Proliferation Assay
(Cat.# G3580, G3581,
G3582) provides an increased level of convenience by combining the novel tetrazolium compound
MTS*, [3-(4, 5-dimethylthiazol-2-yl)-5-(3 carboxymethoxyphenyl)-2-(
4-sulfophenyl)-2H-tetrazolium, inner salt], and the electron coupling reagent phenazine
ethosulfate (PES). The CellTiter 96® AQueous One Solution Reagent
is added directly to cells; no pre-mixing of components is needed. Because of its
stability and the fact that the MTS reduction product does not require solubilization, the
CellTiter 96® AQueous Cell Proliferation Assay requires fewer steps
than other cell viability assays that contain tetrazolium compounds (e.g., MTT and INT).
All of the CellTiter 96® AQueous Cell Proliferation Assays are
non-radioactive, colorimetric assays for determining the number of viable cells in
proliferation and cytotoxicity assays. MTS (Owens reagent) is bioreduced by cells
into a colored formazan product that is soluble in tissue culture medium. The convenient
One Solution format represents an improvement over the current CellTiter 96®
AQueous Non-Radioactive Cell Proliferation Assay where phenazine methosulfate
(PMS) is used as the electron coupling reagent, and solutions of PMS and MTS are supplied
separately.
CellTiter 96® AQueous MTS Reagent Powder
CellTiter 96® AQueous MTS Reagent Powder (Cat.#
G1111, G1112) is now available in
250mg for those who perform intermediate scale proliferation assays. MTS is the
tetrazolium dye found in all CellTiter 96® AQueous Non-Radioactive
Cell Proliferation Assays. Colorimetric measurement of cellular proliferation is monitored
by the conversion of MTS into a soluble formazan product. PMS (phenazine methosulfate) is
not supplied and must be purchased separately. For more information on the use of this

Emax® ImmunoAssay
Systems

Over the past year, Promega has introduced five systems featuring the robust
methodology of the enzyme immunoassay (EIA). These systems feature specific mono- and
polyclonal antibodies for the detection of soluble glial cell-line derived neurotrophic
factor (GDNF; Cat.# G7620,
G7621), nerve growth factor (NGF; Cat.#
G7630, G7631) (introduced in this issue), neurotrophin-3 (NT-3; Cat.#
G7640, G7641)
and transforming growth factors (TGFs) beta1 (Cat.#
G7590, G7591) and beta2 (Cat.#
G7600).
NGF Emax® ImmunoAssay
Systems*
Promega recently developed the NGF Emax ImmunoAssay System (Cat.#
G7630, G7631) for the sensitive detection of NGF in a variety of
neuroscience research applications. This system allows accurate
quantitation of NGF in serum or tissue homogenates and can detect NGF in many
mammalian species. These systems contain sufficient reagents for 160 or 400
determinations of experimental samples, plus human NGF standards in an antibody
sandwich format.
GDNF Emax® ImmunoAssay

The GDNF Emax® ImmunoAssay System (Cat.#
G7620, G7621) is now
available in a convenient 2 x 96 well plate size for those who perform fewer assays, or
for those desiring to test the system for their specific application in small pilot
trials. This system allows accurate quantitation of GDNF in the range of 16-1,000pg/ml.
The specificity of the GDNF EmaxTM ImmunoAssay
Systems were tested with recombinant human NGF and TGFalpha, and natural human, TGFbeta1,
which showed less than 2% cross-reactivity.
Nerve Growth Factor, 2.5S, Murine
Promega now offers its popular mNGF, 2.5S, in a convenient 10µg size (Cat.#
G5142) for those
requiring smaller quantities of this important growth factor. Promegas 2.5S mNGF,
supplied lyophilized, exhibits an ED50 value below 2ng/ml using a PC12 neurite outgrowth
assay.
