Technical Evaluation of Four STRs Multiplex Kits: AmpFlSTRÔ Blue and Green (by Perkin-Elmer), GenePrintÔ CTTv (by Promega) and SGM (FSS) in Italian Forensic Caseworks

Currently, the forensic DNA community analyses DNA extracted from biological evidence by PCR using commercial kits like AmpliType®PM (PolyMarker) (1) based on the reverse dot blot typing technology or/and STRs Multiplex Systems using fluorescent detection systems.
This study was designed to evaluate the amplification efficiency of DNA extracted from bloodstains, hairs and cigarette butts using different commercial kits in the same condition starting. DNA extracted by Chelex method (2) was quantified using QuantiBlot kit by P.E. and the same quantity of DNA (1-2.5 ng) was amplified following the protocols described in the Users Manual. All the samples were amplified in the GeneAmp PCR Systems 9600 (PE). PCR products were visualized with ABI PRISMÔ 377 DNA Sequencer.
The study shows that it is difficult to set the condition of PCR from forensic samples, in fact the results indicate that there are significant difference of PCR efficiency between the several kits.
