Polymorphisms of STR Myelin Basic Protein Locus in Chinese

STR myelin basic protein (MBP) gene showing a high degree of polymorphism, that two system in DNA typing can be amplified by PCR with one pair of primers (Boylan et al., 1990). The allele and genotype of MBP locus were analyzed in 205 unrelated Chinese. Analysis of amplified DNA samples were separated by 6.5% polyacrylamide gel electrophoresis and visualized by silver staining. A total of 13 alleles in high molecular weight region (A) and 6 alleles in low molecular weight region (B) of STR MBP were found, moreover, 43 genotypes of MBP-A and 16 genotypes of MBP-B were observed. Four common alleles for MBP-A were allele 10, 12, 13, and 14, and for MBP-B were allele 7, 9, 10 and 11. The polymorphism information content (PIC) in this population was calculated as 0.85 for MBP A and 0.72 for MBP-B. The power of discrimination (PD) was 0.96 for MBP-A and 0.90 for MBP-B. No significant deviation from Hardy-Weinberg equilibrium could be observed for this study.
