Validation of Six STR Loci for Case Work

Short Tandem Repeats (STRs) are loci that typically contain a series of 2-6 bp tandem repeats. We have completed validation work on the four Home Office tetranucleotide loci: VWA, HUMTH01, F13A1 and FES/FPS, and two additional loci: CSF and TPOX. Individual fluorescent primer sets for these four loci were released by ABD (Applied Biosystems Division of Perkin Elmer) in August, 1995, which we have combined in a multiplex PCR.

Prior to use of these systems in casework, a number of studies were undertaken in order to validate these systems. These include population studies, sensitivity studies, mixture studies, intra- and inter-gel sizing comparisons and the analysis of casework samples which had previously been characterized with other DNA loci. Results of these studies will be presented.
