We have developed a series of multiplex STR systems for rapid, high throughput, nonisotopic, and inexpensive DNA analyses. We have achieved high throughput analysis with STR loci for forensic analysis and paternity determination using manual separation and silver stain detection to avoid the need for specialized or expensive equipment.
Nine STR loci have been incorporated into two multiplexes, each containing three loci. Six of these loci are included in two triplexes, CSF1PO, TPOX, TH01, (CTT), and F13A01, FESFPS, and vWA (FFv) described in a recent journal article.
Recently, we have developed the GenePrinttm SilverSTRtm III System containing the STR loci D16S539, D7S820, D13S317. A survey of over 200 unrelated individuals in each of three racial/ethnic groups indicates that this triplex is the most polymorphic of the set in both Caucasian-American and Hispanic-American populations, but shows more moderate polymorphism among African-Americans.

The use of all three GenePrinttm SilverSTRtm Systems (i.e., CTT, FFv, and SilverSTRtm III) provides enormous discrimination power from the nine STR loci D16S539, D7S820, D13S317. A survey of over 200 unrelated individuals in each of three racial/ethnic groups indicates that this triplex is the most polymorphic of the set in both Caucasian-American and Hispanic-American populations, but shows more moderate polymorphism
among African-Americans.

With Typical Paternity Indices exceeding 500 in each racial/ethnic group, silver stain multiplex STR systems now have the discrimination power to be used in this application.
