Background-Free Chemiluminscent Detection on Biodyne B Membranes Using LifeCodes Quick-Light System

In order to expedite forensic RFLP detection, the St. Louis Metropolitan Police Department Crime Laboratory converted from 32P to chemiluminescent detection. We had been using an FBI protocol which included Southern blotting onto a charged membrane (Biodyne B) followed by 32P detection. Each probing required from a few days to a few weeks. Conversion to chemiluminescent detection considerably shortened detection time to hours. Establishing a chemiluminescent protocol while retaining the use of Biodyne B membranes facilitated the change-over. Both newly-produced membranes and older, previously 32P probed membranes could undergo additional probings together. One protocol does it all!
