Which DNA polymerase is preferred for removal of a 3´ DNA overhang or for filling in a 5´ overhang?

While both T4 DNA Polymerase (Cat.# M4211) and DNA Polymerase I Large (Klenow) Fragment (Cat.# M2201) have a 3´→ 5´ exonuclease activity that can be used to enzymatically remove a 3´ overhang, T4 DNA Polymerase is often the preferred enzyme. T4 DNA Polymerase has a stronger 3´→ 5´ exonuclease activity that will remove the overhang more efficiently. Both enzymes can be used to fill in a 5´ overhang, and there is not often a preference between these enzymes. The removal of a 3´ overhang with T4 DNA Polymerase requires the addition of dNTPs.
