RNAi Knockdown in Primary Cells: Suppression of Target Gene with psiSTRIKE™ and psiCHECK™ Vectors and amaxa's Nucleofector® Technology

Efficient delivery of highly effective shRNAs into primary cells enables studying gene function in a biologically relevant context. Here we provide an example of successful gene knockdown in primary cells using an RNAi approach. Effective shRNAs were rapidly screened and selected using the psiCHECK™-2 Vector. Small hairpin interfering RNAs were expressed from psiSTRIKE™–hMGFP Vectors that were efficiently delivered into primary cells using amaxa’s Nucleofector® Technology.
