Notes:
In an attempt to understand the signaling mechanisms involved in neurotrophic regulation of synaptic transmission at the neuromuscular junction, the authors examine the phosphoinositide-3 kinase, phospholipase C, and MAP kinase signaling pathways. The levels of Akt-1 phosphorlyation was determined by immunocytochemistry. Xenopus nerve–muscle cultures were treated with or without various kinase inhibitorsand stimulated with neurotrophin-3. The cells were fixed with 3% paraformaldehyde and 0.25 glutaraldehyde, permeabilized with 0.25% Triton® X-100, and incubated with the Anti-pS473 Akt pAb (1:100 dilution).
