Sexual assault samples provide valuable forensic information. However, mixture samples are among the most difficult to process, as sperm and epithelial fractions must first be separated. The automated Differex™ System provides an alternative to cumbersome manual methods by introducing automation into the differential extraction process. Robotic processing of mixture samples makes it possible to reduce case backlogs without compromising quality.
The automated Differex System combines Differex reagents with proven DNA IQ chemistry and specialized methods to achieve complete and clean differential extraction results.
Figure 1. The automated Differex™ method uses DNA IQ™ Resin to cap the sperm pellet after differential lysis and centrifugation. The MagnaBot® Flat Top Magnetic Separation Device immobilizes the DNA IQ resin, holding the sperm pellet in place during automated wash steps. The Separation Solution helps, remove any remaining digestion buffer and epithelial DNA away from the sperm pellet.
The Power of Speed
It can take up to 5 days to process 40 differential extraction samples using the typical manual differential extraction method, and the final results may be variable depending on the analyst's skill level. With the automated Differex method you can process up to 40 differential extraction samples, from incubation to final eluted DNA, in less than 5 hours. Hands-on time is less than 1 hour, reducing total processing time by up to 90%!
