What is a ddRNAi expression cassette?
The ddRNAi expression cassette is a short fragment of DNA (300bp) that contains the U6 promoter sequence. Through amplification the siRNA target sequence is introduced into the cassette. The cassette is then directly transfected into a mammalian cell or cloned into the appropriate vector.

What is the primary use of a ddRNAi expression cassette?
Although some algorithms are designed to predict how effective a given siRNA will be at suppressing the expression of its target, selecting optimal regions still remains largely a trial-and-error process. Therefore, multiple regions are generally screened for each target. Screening can be performed using synthetic RNAs, which are expensive. The use of ddRNAi expression cassettes allow for an inexpensive and easy format for screening several siRNA target sequences.
