The MagneHis Protein Purification System provides a simple, rapid and reliable method for the purification of polyhistidine- or HQ-tagged, expressed proteins. Paramagnetic precharged nickel particles (MagneHis Ni-Particles) are used to isolate polyhistidine- or HQ-tagged proteins directly from a crude lysate of bacterial, insect or mammalian cells, or from culture medium using either a manual or automated procedure.
Bacterial, insect or mammalian cells containing a polyhistidine- or HQ-tagged protein are lysed directly in their culture medium using the FastBreak™ Cell Lysis Reagent, 10X and MagneHis Ni-Particles are added to the lysate. The polyhistidine- or HQ-tagged protein binds to the Ni-Particles in a matter of minutes. The unbound protein is washed away and the target protein is recovered by elution with imidazole.
