Wizard® SV 96 Genomic DNA Purification System

The Wizard® SV 96 Genomic DNA Purification System provides a fast, simple technique for the preparation of genomic DNA from cultured cells and tissue, including mouse tails. Genomic DNA can be purified from cultured cells in about 30 minutes and isolation of tissue or mouse tails in less than one hour, with an overnight digest with Proteinase K. Amplifiable genomic DNA can be isolated from up to 5 × 106 cells per prep, from 20mg of tissue or from up to 1.2cm of a mouse tail tip without a centrifugation clearing step prior to purification.

With the Wizard® SV 96 Genomic DNA Purification System, genomic DNA is purified from cell lysates using 96-well vacuum filtration, eliminating the need for centrifugation. Purification requires the use of the Vac-Man® 96 Vacuum Manifold or similar 96-well manifold. The technique also provides the ability for automation on liquid handling workstations such as the Beckman Biomek® 2000. 

High-throughput genomic DNA purification is made easy because the SV 96 System uses a convenient vacuum manifold apparatus that does not require disassembly during the genomic DNA binding and wash steps. Filtrate waste products are delivered directly to a vacuum trap, eliminating the need to empty waste collection vessels. Purified genomic DNA is eluted into a 96-well deep-well plate.

FEATURES

Fast: Obtain genomic DNA from 96 mouse tails in 45-60 minutes, cultured cells in 30 minutes after lysis. No spin required.

Flexible: Purify genomic DNA from up to 20mg of tissue, 1.2cm of mouse tail or 5 x 106 cultured cells per well. Tissue isolations require Proteinase K.

PCR products were amplified from 1µl of purified sample from each well using primers for mouse IL-1ß. Ten microliters of PCR product were separated on a 1% gel and visualized by ethidium bromide staining. Expected IL-1ß PCR product from mouse tail clippings is approximately 1.2kb. No PCR product was expected from water samples. Reactions were amplified for 30 cycles.

High Yield: Purify 20-30µg of DNA per prep from 1.2cm mouse tail clips.
