PowerPlex® 2.1 System

Description

The PowerPlex® 2.1 System allows the co-amplification and two-color detection of nine STR loci. The PowerPlex® 2.1 System contains loci Penta E, D18S51, D21S11, TH01, D3S1358, FGA, TPOX, D8S1179 and vWA. In the PowerPlex® 2.1 System, one of the two primers for Penta E, D18S51, D21S11, TH01 and D3S1358 is labeled with fluorescein (FL), and one primer specific for FGA, TPOX, D8S1179 and vWA is labeled with carboxy-tetramethyl-rhodamine (TMR). All nine loci are amplified simultaneously in a single tube and analyzed in a single gel lane.

The PowerPlex® 2.1 System is customized for use with the Hitachi FMBIO® and Hitachi FMBIO® II Fluorescence Imaging Systems.

Features

  • High-Throughput Analysis:

  • Analysis is achieved by comparison of amplified sample DNA fragments directly with the allelic ladder present for each locus.
  • Internal Lane Standard 600:

  • This marker (Cat.# DG2611) offers the greatest precision available for DNA typing. It is designed for use in each gel lane to increase the precision of your analyses. The CXR-labeled marker consists of 22 bands ranging in size from 60–600bp. Fragments that are multiples of 100 bases have fluorescence intensities that are approximately twice that of other fragments to simplify size assignment.
  • Allelic Ladders Mix:

  • Allelic ladders for all 16 loci, labeled with three fluorescent dyes, are provided to assure balance across loci and simplify interpretation of alleles.
  • Internal Lane Standard (ILS) 600 BIO:

  • The marker is designed for use in each gel lane to increase precision in analyses. The ILS 600 BIO contains 21 evenly balanced fragments ranging from 80–600bp.
  • Allelic Ladders:

  • Allelic ladders for all loci are provided to assure balance across loci. This simplifies interpretation and improves software compatibility.
  • Penta E:

  • The low level of stutter associated with Penta E simplifies the interpretation of results from samples containing more than one source of human DNA. In addition, it is a highly polymorphic locus and has no known microvariant alleles. These combined properties make the Penta E locus an ideal marker for forensic analysis.
  • Efficient:

  • An allelic ladder mixture consisting of most or all of the known alleles in the population is included to allow rapid and precise assignment of sample alleles. Less than one day required for sample to data analysis.
  • Sensitive:

  • Amplification of 1–250ng of genomic DNA.
  • Tolerant:

  • More tolerant of degraded DNA than other typing methods.
