Description
The PowerPlex® 16 System is a multiplex STR system for use in DNA typing,
including paternity testing, forensic DNA analysis, human identity testing
and tissue culture strain identification. The System allows the coamplification
and three-color detection of sixteen loci (fifteen STR loci and Amelogenin).
The PowerPlex® 16 System contains the loci Penta E, D18S51, D21S11, TH01, D3S1358,
FGA, TPOX, D8S1179, vWA, Amelogenin, Penta D, CSF1PO, D16S539, D7S820, D13S317
and D5S818. All sixteen loci are amplified simultaneously in a single tube and
analyzed in a single injection or gel lane.
The loci included in the PowerPlex® 16 System have been selected because they
satisfy the needs of several major standardization bodies throughout the world,
including the European police network, INTERPOL, the European Network of
Forensic Science Institutes (ENFSI), GITAD (Grupo Iberoamericano de Trabajo
en Análisis de DNA) and the United States Federal Bureau of Investigation (FBI).
The system has undergone extensive validation efforts, including external CODIS
validation for database and forensic casework, as well as internal validation.
The PowerPlex® 16 System also contains two new low-stutter, highly polymorphic
pentanucleotide repeat loci, Penta E and Penta D. These loci add significantly
to the discrimination power of the system, making the PowerPlex® 16 System a
single amplification system with a power of exclusion sufficient to resolve
paternity disputes definitively. In addition, the extremely low stutter seen
with Penta E and Penta D, makes them ideal loci for evaluation of DNA mixtures
often encountered in forensic casework.
Finally, the Amelogenin locus is included in the PowerPlex® 16 System to allow
gender identification of each sample.
The PowerPlex 16 System is designed specifically for use with the Applied Biosystems 3130 and 3130xl Genetic Analyzers, the ABI PRISM 3100 and 3100-Avant Genetic Analyzers, and the ABI PRISM 310 Genetic Analyzer.
Matrix generation is performed using the PowerPlex Matrix Standards, 310 (DG4640) or PowerPlex Matrix Standards, 3100/3130 (DG4650).

Amplify DNA Once, Not Twice.
- No longer do you have to amplify each database or casework sample twice.
With the PowerPlex® 16 System you can amplify all 13 CODIS STR loci at one time.
Get the sensitivity you require.
- The PowerPlex® 16 System provides maximum sensitivity with low quantities of
DNA—it is optimized for use with 1ng of DNA. However, each lot is
performance tested to provide reproducible results with 0.5ng of DNA.
Additional studies show interpretable results can be obtained with <0.5ng of DNA.

Concordance.
- PowerPlex® 16 is concordant with Applied Biosystem’s Profiler Plus™, COfiler™
and Identifiler™ PCR Amplification kits.
Validation Studies.
- Promega’s developmental validation was completed in October 2000.
In 2001, more than 20 laboratories participated in a casework validation study
to comply with SWGDAM guidelines. Over 2000 samples were typed by multiple
CODIS laboratories the required CODIS database validation. Results of these
studies are published and are available upon request.
Realize the speed and simplicity you want.
Primer Set.
- Consistent balance across loci and between dyes simplifies analysis of low amounts of
template DNA and minor contributors of other DNA samples.
- This CXR-labeled marker offers the greatest precision available for DNA typing.
Peaks range in size from 60–600bp. Fragments that are multiples of 100 bases
are present at approximately twice the height of other fragments to simplify
size assignment.
Allelic Ladders.
- Allelic Ladders for all 16 loci are provided to
simplify interpretation and improve software compatibility.
PowerTyper™ Macros.
- The PowerTyper™ Macros has been designed to automatically label fragments
generated from GeneScan® data.
