PowerPlex® 1.2 System

Description

The PowerPlex® 1.2 System provides a rapid, non-radioactive method designed to obtain a DNA type, or genetic fingerprint using very small amounts (e.g., 1ng) of human DNA. The PowerPlex® 1.2 System is designed specifically for use with the ABI PRISM® 310 Genetic Analyzer and is also compatible with the ABI PRISM® 373 and 377 DNA Sequencers. PowerPlex® 1.2 allows for the simultaneous single-tube amplification and two-color detection of eight polymorphic STR loci. CSF1PO, TPOX, TH01 and vWA primers are labeled with tetramethylrhodamine; D16S539, D7S820, D13S317 and D5S818 primers are labeled with fluorescein. All of the loci included in this set are true tetranucleotide repeats. To facilitate analysis of the data generated with the PowerPlex® 1.2 System, we have created a file to allow automatic assignment of genotypes using the ABI Genotyper® Software. The PowerTyper™ 1.2 Macro is supplied on disk and facilitates analyzing the PowerPlex® 1.2 data. The PowerTyper™ 1.2 Macro is available upon request at no additional charge and is also available online.

Features

  • High-Throughput Analysis:

  • Analysis is achieved by comparison of amplified sample DNA fragments directly with the allelic ladder present for each locus.
  • Size Marker:

  • Fluorescent Ladder (CXR), 60–400 Bases, is provided with the system and is tagged with a third fluorescent dye. This marker may be included as an internal lane standard in each lane to increase precision in analyses when using the PowerPlex® 1.2 System.
  • Allelic Ladders:

  • Allelic ladders for all loci are provided to assure balance across loci. This simplifies interpretation and improves software compatibility.
  • Efficient:

  • The PowerPlex® 1.2 System allows simultaneous amplification and analysis of the eight STR loci plus Amelogenin for gender identification. The Amelogenin system allows amplification of sequences specific to the X (212-base) and Y (218-base) chromosomes, respectively.
  • Sensitive:

  • Amplification of 1–250ng of genomic DNA.
  • Tolerant:

  • More tolerant of degraded DNA than other typing methods.
