PowerPlex® 1.1 System

Description

The PowerPlex® 1.1 System provides a rapid, non-radioactive method designed to obtain a DNA type, or genetic fingerprint, using very small amounts (e.g., 1ng) of human DNA. The PowerPlex® 1.1 System allows the co-amplification and two-color detection of eight STR loci. CSFIPO, TPOX, TH0I and vWA primers are labeled with tetramethylrhodamine; D16S539, D7S820, D13S317 and D5S818 primers are labeled with fluorescein. All eight loci are amplified simultaneously in a single tube and analyzed in a single gel lane. Loci amplified using the PowerPlex® 1.1 System also may be co-amplified with the Fluorescent Sex Identification System-Amelogenin (TMR), allowing the detection of nine loci in a single gel lane. The PowerPlex® 1.1 System is customized for use with the Hitachi FMBIO® and Hitachi FMBIO® II Fluorescence Imaging Systems.

Features

  • High-Throughput Analysis:

  • Analysis is achieved by comparison of amplified sample DNA fragments directly with the allelic ladder present for each locus.
  • Size Marker:

  • Fluorescent Ladder (CXR), 60–400 Bases, is provided with the system. It is tagged with a third fluorescent dye. This marker may be included as a standard in each gel lane and visualized separately to monitor fluctuations in sample mobility that may occur from lane-to-lane.
  • Allelic Ladders:

  • Allelic ladders for all loci are provided to assure balance across loci. This simplifies interpretation and improves software compatibility.
  • Flexible:

  • The primer sets are compatible with use of either AmpliTaq Gold® (when used with Gold ST*R 10X Buffer [Cat.# DM2411] sold separately) or more generic forms of Taq DNA polymerase, generating few or no amplification artifacts with either.
  • Sensitive:

  • Amplification of 1–250ng of genomic DNA.
  • Tolerant:

  • More tolerant of degraded DNA than other typing methods.
