DNA IQ™ System

Description

The DNA IQ™ System is a DNA isolation and quantitation system designed specifically for the forensic and paternity community. This system employs a novel technology with magnetic particles to prepare clean samples for short tandem repeat (STR) analysis easily and efficiently. The DNA IQ™ System can be used to extract DNA from stains or liquid samples such as blood or resulting DNA solutions.

The unique DNA IQ™ Resin is designed to eliminate PCR inhibitors and purify contamination frequently encountered in casework samples. When working with larger samples found in paternity and databasing, the DNA IQ™ System automatically delivers a consistent amount of total DNA. Buccal swabs, stains on FTA® paper and liquid blood have been analyzed with the system (the DNA IQ™ Spin Baskets are not required for liquid blood analysis).

The DNA IQ™ System has been tested with the AluQuant® Human DNA Quantitation System and the PowerPlex® 16 System to ensure a streamlined process. This translates into reliable products giving optimal results from isolation to quantitation and STR analysis.

  • Designed specifically for use on forensic, database and paternity samples:

  • When working with small amounts of DNA, the system becomes more efficient as sample size decreases. The unique DNA IQ™ Resin used in the system is designed to eliminate PCR inhibitors and purify the contaminated samples frequently encountered in casework (e.g. denim, leather, and soil). DNA IQ™ can be used to analyze difficult samples such as bloodstains on denim. Even bloodstains as small as 0.1µl can be isolated and amplified to give a complete STR profile!.
  • No extra quantitation steps prior to amplification:

  • For paternity and database samples, DNA IQ™ delivers a consistent amount of total DNA. It doesn't matter if the samples are being isolated from swabs, stains on FTA® paper, or liquid blood, DNA IQ™ will deliver consistent yields of pure DNA ready for amplification without any added quantitation steps.
  • Cleaner DNA with a simple, rapid system:

  • Compared to other systems, DNA IQ™ makes it easier to get from sample to cleaner, quantitated DNA. To extract stains, just heat your sample in the Lysis Buffer for 30 minutes and centrifuge. DNA IQ™ Resin is then added to the solution and the samples are washed using a magnetic stand. Thirty minutes later, you have purified DNA.
  • Flexible:

  • The DNA IQ™ System is flexible. One simple system can be used for all types of forensic samples. The DNA IQ™ System allows you to decide how much DNA to extract from your samples.
  • Efficient:

  • Sensitive to minute sample sizes. In addition, no harmful, organic solvents such as phenol and chloroform are used, so use of a hood is not required and disposal of hazardous chemicals is eliminated.

Figure 1. 0.1μl Bloodstains. Blood was diluted 10 fold with PBS and 1 μl spots (0.1μl blood) were placed on blue denim and stored for at least 3 weeks at room temperature. These 0.1μl bloodstains were then cut out, eluted and purified using 25μl elution volumes. 5μl out of 25μl of eluted DNA were placed in PowerPlex® 16 reactions and analyzed on an ABI PRISM® 310 capillary electrophoresis system. All samples examined gave easily identified genotypes.

Figure 2. Buccal Swab Application. Buccal swabs (cheek scrapings) obtain variable amounts of DNA (200 to 3,000ng) from individual to individual and normally require a quantitation step prior to PCR amplification. The DNA IQ™ System eliminates the need for this quantitation step.

Buccal swabs from 12 individuals were obtained, extracted, and purified with the DNA IQ™ System. 1 μl of eluted DNA was amplified with PowerPlex® 1.1 and analyzed on an FMBIO® II.

Genotypes were easily obtained for all samples
