MagneSil® ONE, Fixed Yield 
Blood Genomic System

If you are in large-scale clinical or academic genotyping research one of your major concerns is most likely sample preparation. You need to purify DNA from a large number of blood or other samples but are spending too much time performing quantitation and normalization of each DNA sample prior to amplification.  What you need is speed, but what you really need is a walk-away method that normalizes DNA yield during purification.  The MagneSil® ONE, Fixed Yield Blood Genomic System frees up your time.

The MagneSil® ONE, Fixed Yield Blood Genomic System is a high-throughput system for DNA purification from blood. The system is designed to purify a “fixed yield” of 1µg of DNA (+/-50%) from 50-60ul of anti-coagulated liquid blood in a 96-well walk-away automated format. Set up and place the blood samples on the instrument and start the instrument. That’s it. A Beckman Coulter Biomek® FX robot will complete purification in about 1 hour.

Example yield from 12 blood donors, 4 replicated samples. Average yield is 919ng with an average A260/A280 ratio of 1.89.

Purification is accomplished using Promega’s MagneSil®- Fixed Yield paramagnetic particles, optimized buffers, and the Deep Well MagnaBot® Magnetic Separation Stand. The procedure consists of three simple steps. 

  1. Whole blood is added to Lysis Buffer and MagneSil particles and is mixed to lyse white cells and bind DNA to the particles. 
  2. Washes are done to remove heme, protein and other contaminants. 
  3. Finally, a fixed yield of DNA is eluted in Elution Buffer ready to be analyzed by PCR, multiplex PCR, or other genotyping applications.
