Wizard® MagneSil™ Sequencing Reaction Clean-Up System

The Wizard® MagneSil™ Sequencing Reaction Clean-Up System is a  high-throughput solution to sample purification. The system is designed to purify BigDye™ terminator cycle sequencing reactions to ensure optimal fluorescent sequencing results. The system removes buffer salts and unincorporated dye-labeled terminators from sequencing extension products prior to loading onto a fluorescent instrument.  The procedure can be performed in a standard 96-well PCR plate and may be processed on a robotic workstation.

Clean up is peformed using Promega's unique MagneSil™ GREEN paramagnetic particles and the MagnaBot® II Magnetic Separation Device.  This innovative device is designed to work with most robotic platforms. 

The procedure consists of an easily automated three-step protocol.   MagneSil™ particles selectively bind to sequencing extension products.   Non-specifically adsorbed material is washed away with ethanol.  The purified sequencing extension products are eluted in gel loading solution and are ready to be analyzed by high-throughput DNA sequencing.

Qualified for Fluorescent Sequencing

MagneSil™ systems undergo functional testing to ensure performance in fluorescent capillary and gel electrophoresis sequencing systems. The system is qualified to produce a lock on position of less than or equal to 25 bases.

Consistent Performance

Sequencing data quality meets or exceeds that obtained with gel filtration or ethanol precipitation methods.

Reproducible Results

Typically, Phred 20 values for sequencing reactions purified with the MagneSil™ Sequencing Reaction Clean-Up System are greater than or equal to 600 bases.

The MagneSil™ Sequencing Reaction Clean-Up System offers the distinct advantage of a hands-off protocol while achieving sequencing quality that is superior to reactions purified by gel filtration or ethanol precipitation.  The system undergoes rigorous testing for performance to ensure it can meet or exceed 98% accuracy out to 600 bases of sequence and routinely achieves 98% accuracy out to 700 bases.
