Wizard® MagneSil Plasmid Purification
System
The Wizard®
MagneSil Plasmid Purification System offers a cost-effective method for purifying
high-quality plasmid DNA. The system is designed exclusively for use with automated
instruments allowing truly "walkaway" high-throughput plasmid purification.
DNA isolation is performed using
Promega's unique MagneSil Paramagnetic Particles and a magnetic separation device,
such as the MagnaBot® 96 well Magnetic Separation Device, which is adaptable to any
robotic platform. The protocol has also been optimized using an orbital shaker, such
as the Micro Mix 5 orbital shaker with fitted platform and plate holders
(Cat.# G5016 MM5
kit, Acme Automation, www.acme-automation.com).
MagneSil Paramagnetic Particles are used in
separate steps to clear bacterial lysates (MagneSil BLUE) and to capture plasmid DNA
(MagneSil RED). The use of particles circumvents the need for centrifugation or
vacuum manifolds, making the system ideal for automation. Eluted DNA is ready for use in
fluorescent sequencing reactions as well as other standard molecular biology
techniques. Protocols are available for
plasmid purification in a 96 well format.

The Wizard® MagneSil Plasmid Purification System purifies high-quality plasmid DNA for use in a number of molecular biology applications including
fluorescent sequencing. The system undergoes rigorous testing for performance in dye
terminator sequencing to ensure it can meet or exceed 600 bases with greater than 98%
accuracy.

Consistent Recovery
Reliable recovery of plasmid DNA is ensured. A number of factors including plasmid copy number and cell
density affect the final yield of plasmid DNA. Another important factor is how
consistent the system is at purifying multiple samples. The MagneSil Plasmid
Purification System has been tested at a number of cell densities to ensure that DNA
recovery is reliable across multiple wells of a 96 well plate as well as between plates.

Reproducible Results
MagneSil products can be relied upon to purify high-quality template for fluorescent
sequencing. High-throughput sequencing requires a
reliable purification system. The Wizard® MagneSil Plasmid Purification System can be
relied on to produce high-quality templates time and again to ensure accurate sequencing.
