Characteristic of microsatellites instability is abnormal variation of repetitive nucleotide sequences in DNA from the tumor compared to DNA of healthy tissue from the same individual.
 

Determining whether a tumor exhibits microsatellite instability (MSI) is useful in identifying patients with hereditary non-polyposis colorectal cancer (HNPCC) and sporadic gastrointestinal cancers with defective mismatch repair system (MMR). There is evidence to suggest that colorectal cancer with MSI-H respond differently to adjuvant chemotherapy so the assessment of MSI-H status aids in establishing a clinical prognosis.
 

Promega has developed, based on previous studies done by Dr Hamelin and his group, the MSI analysis system. This fluorescent multiplex PCR system meets the 2002 NCI workshop recommendations and includes five mononucleotides markers (BAT-25, BAT-26, NR-21, NR-24 and MONO-27). In addition two pentanucleotide markers were added to identify sample mixs-up and/or contamination. PCR products are separated by capillary electrophoresis. A MSI analysis macro software simplifies and standardizes allele size determination for consistent data analysis.

The total procedure from DNA extraction to MSI analysis will be described during the presentation.
